The first test that was performed was a streak plate on Tryptic Soy Agar (TSA). The streak plate method produces isolated colonies on the agar plate;, allowing for the observation of a bacteriasbacteria's individual colony morphology. Colony morphology can aid in identification because each bacteria produces different-looking colonies. There are a few basic elements that can be used to identify bacterial colonies;, including color, shape, and elevation. When examined on the TSA streak plate, the unknown bacterial colonies were yellow, convex, pinheaded, and circular in shape. After the streak plate was performed, the bacteria was inoculated on a Mannitol Salt Agar plate (MSA). MSA is a selective and differential medium, it. It contains high levels of saline, as well as the sugar mannitol and a pH indicator known as phenol red. If growth occurs, then it is able to be determined ifenables determination of whether the bacteria is halophilic and if it can ferment mannitol. If the bacteria is capable of mannitol fermentation or can survive in high salt concentrations, then the phenol red in the agar will turn yellow. This is due to an acidic byproduct that is produced during fermentation. After examining the plate for bacteria 2A it was determined that the organism was capable of mannitol fermentation and was able to survive in the presence of high salt concentrations.
